QuEchERS Products
QuEChERS was first proposed at EPRW conference in 2002, and aimed to establish a quick, cheap pretreatment method for multi residue pesticide analysis in fruits, vegetables, grains and lowfat products. Specifically, solid-phase extraction adsorbents are dispersed in sample extractions to adsorb interference and retain analytes, and then the purified sample can be analysed directly by chromatography. Such as the abbreviation of "QuEChERS"(Quick, Easy, Cheap, Effective, Rugged and Safe), this method is Quick, Easy, Cheap, Effective, Rugged and Safe. QuEChERS is approved by AOAC and EN European Pesticide Residue Monitoring
Committee. Compared QuEChERS with the traditional method extensively used throughout Europe for 15–20 years and calculated an approximate 95% reduction in solvent consumption (10 versus 535 mL), an approximate 95% reduction in consumable costs, and an approximate 90% reduction in time.
Introduction of QuEChERS
EN 15662 method
The original QuEChERS method is non-buffered, in order to extract some pH sensitive compounds and decrease the pesticide decomposition, a certain amount of buffer is added into the extraction tube to expand the detection range. But now the two popular method AOAC 2007.1 and EN 15662 introduce NaAcetate or NaCitrate to be the buffer. Based on different matrix's pH and different types of interferences, we have special extraction tubes and clean up tubes as below:
Add 15g Comminuted sample in to 50ml centrifuge tube with cap, add 15mL 1% acetic acid ACETONITRILE and 100ul internal standard, shake 1min
Add 6g MgSO4 1.5g NaAcetate(AOAC 2007.01 centrifuge tube for sample extraction) shake 1min, 4000r/min centrifuge 5min
Judged by the content of pigment, fatty and waxy in the matrix, collect different AOAC 2007.01 clean-up centrifuge tubes, add the upper layer liquid, shake 30s, 4000r/min centrifuge 5min
AOAC 2007 method
1add 10g Comminuted sample in to 50ml centrifuge tube with cap, add 10ml ACETONITRILE (or 1% acetic acid ACETONITRILE) and 100ul internal standard, shake 1min
Add MgSO4,NaCl, buffer(NaAcetate or NaCitrate) ,adjust PH to 5 , most pesticide are stable in this condition, shake 1min, 4000r/min centrifuge 5min
Add PSA(remove fatty acid) GCB(remove pigment) and MgSO4(remove water), shake 1min, 4000r/min centrifuge 5min
GC, LC or GC/MS LC/MS
Add 10g Comminuted sample in to 50ml centrifuge tube with cap, add 10ml ACETONITRILE and 100ul internal standard, shake 1min
Add 4g MgSO4 1g Nacl 0.5g disodium citrate Sesquihydrate, 1g NaCitrate (EN 15662 centrifuge tube for sample extraction) shake 1min, 4000r/min centrifuge 5min
Judged by the content of pigment, fatty and waxy in the matrix, collect different EN 15662clean-up centrifuge tubes, add the upper layer liquid, shake 30s, 4000r/min centrifuge 5min
